De Lck HTRF kinase-assay omvat ATP-afhankelijke fosforylering van een gebiotinyleerd substraatpeptide van gastrine in aanwezigheid of afwezigheid van een remmerverbinding. De uiteindelijke concentratie van gastrine is 1,2 μM. De uiteindelijke concentratie van ATP is 0,5 μM (Km app = 0,6 ± 0,1 μM), en de uiteindelijke concentratie van Lck (een GST-kinase-domeinfusie (AA 225−509)) is 250 pM. Buffercondities zijn als volgt: 50 mM HEPES pH = 7,5, 50 mM NaCl, 20 mM MgCl2, 5 mM MnCl, 2 mM DTT, 0,05% BSA. De assay wordt geblust en gestopt met 160 μL detectiereagens. Detectiereagentia zijn als volgt: Buffer gemaakt van 50 mM Tris, pH = 7,5, 100 mM NaCl, 3 mM EDTA, 0,05% BSA, 0,1% Tween20. Voorafgaand aan het uitlezen wordt streptavidine-allofycocyanine (SA-APC) toegevoegd in een uiteindelijke concentratie in de assay van 0,0004 mg/mL, samen met geëuropileerde anti-fosfotyrosine Ab (Eu-anti-PY) in een uiteindelijke concentratie van 0,025 nM. De assayplaat wordt uitgelezen in een Discovery fluorescentieplaatlezer met excitatie bij 320 nm en emissie bij 615 en 655 nm. Assays voor andere kinasen worden op een vergelijkbare manier uitgevoerd als hierboven beschreven, waarbij de concentraties van enzym, peptisubstraat en ATP die aan de reactie worden toegevoegd, variëren, afhankelijk van de specifieke activiteit van de kinase en de gemeten Kms voor de substraten.
