AZD7762, een meer selectieve Chk1-remmer, remt de Chk1-fosforylering van een cdc25C-peptide door reversibel te binden aan de ATP-bindingsplaats van Chk1, met een IC50 van 5 nM en een Ki van 3,6 nM. Deze verbinding induceert celstop met een EC50 van 0,620 μM, en heft de door camptothecine geïnduceerde G2-arrest significant op met een EC50 van 10 nM, door de Chk1-afhankelijke afbraak van Cdc25A en de activering van Cyclin A te blokkeren. Het (300 nM) verbetert de antitumorale werkzaamheid tegen SW620 en tegen MDA-MB-231 door de GI50-waarden te verlagen van respectievelijk 24,1 nM en 2,25 μM naar 1,08 nM en 0,15 μM. Dit chemische middel vertoont cytotoxiciteit tegen een verscheidenheid aan neuroblastoom-cellijnen met p53 wild type, p53-mutatie, Mdm2-amplificatie of p14-deletie met IC50-waarden variërend van 82,6-505,9 nM. |

Recombinant humaan Chk1 wordt tot expressie gebracht als een S-transferase fusie in insectencellen met behulp van een baculovirus vector en gezuiverd door affiniteitschromatografie. Een synthetisch peptidesubstraat (N-biotinylaminohexanoyl-KKVSRSGLYRSPMPENLNRPR) voor Chk1 wordt gesynthetiseerd. De uiteindelijke testconcentraties van peptide en ATP (koud + 40 nCi [33P]ATP) zijn respectievelijk 0,8 en 1 μM. Verschillende concentraties van deze verbinding, buffer die peptide en Chk1 kinase en ATP bevat, worden achtereenvolgens toegevoegd aan een 384-wells testplaat. De plaat wordt 2 uur geïncubeerd, de reactie wordt gestopt door toevoeging van buffer die EDTA en scintillatieproximiteitsassay-kralen bevat, en de platen worden afgelezen met behulp van een TopCount-lezer. Gegevensanalyse wordt uitgevoerd om een dosisrespons (IC50) te bepalen.

AZD7762 alleen bij 25 mg/kg vertoont weinig antitumorale activiteit bij de H460-DNp53 xenograftmuizen en SW620 xenograftmuizen, maar wanneer het in combinatie wordt toegediend, vertoont deze verbinding een significante antitumorale werkzaamheid bij de twee xenograftmuizen met een log celvernietiging van 0,9 of een behandeld/controlepercentage (%T/C) van 26, zelfs bij een lage dosis van 12,5 mg. De toediening van deze chemische stof in combinatie bij de H460-DNp53 xenograftrat remt het tumorvolume op een dosisafhankelijke manier met %T/C-waarden van 48 en 32 voor respectievelijk 10 en 20 mg/kg. Deze verbinding (25 mg/kg) in combinatie veroorzaakt volledige tumorregressie bij de SW620 xenograftmuizen, waarbij de %T/C significant toeneemt tot respectievelijk -66% en -67%. |

Since DMSO has low toxicity with i.p. injection, hydroxyproplyl-β-cyclodextrin is considered a safer co-solvent in compound formulation. In a previous literature report, 11.3% hydroxyproplyl-β-cyclodextrin was used to formulate it for i.v. injection.
