Staurosporine (STS) is een krachtige PKC-remmer voor PKCα, PKCγ en PKCη met een IC50 van 2 nM, 5 nM en 4 nM, minder krachtig voor PKCδ (20 nM), PKCε (73 nM) en weinig actief voor PKCζ (1086 nM) in celvrije assays. Dit middel vertoont ook remmende activiteiten op andere kinasen, zoals PKA, PKG, S6K, CaMKII, etc. Fase 3.

In vitro
Staurosporine (STS), een microbieel alkaloïde, remt significant proteïne kinase C uit rattenhersenen met een IC50 van 2,7 nM. Het vertoont ook een sterk remmend effect tegen HeLa S3-cellen met een IC50 van 4 nM. Deze verbinding remt een verscheidenheid aan andere proteïnekinasen, waaronder PKA, PKG, fosforylasekinase, S6 Kinase, Myosin light chain kinase (MLCK), CAM PKII, cdc2, v-Src, Lyn, c-Fgr en Syk met een IC50 van respectievelijk 15 nM, 18 nM, 3 nM, 5 nM, 21 nM, 20 nM, 9 nM, 6 nM, 20 nM, 2 nM en 16 nM. Bij 1 μM induceert het >90% apoptose in PC12-cellen. Consistent hiermee induceert deze behandeling een snelle en langdurige verhoging van intracellulaire vrije calciumspiegels [Ca2+]i, accumulatie van mitochondriale reactieve zuurstofsoorten (ROS) en daaropvolgende mitochondriale disfunctie. De apoptose van MCF7-cellen geïnduceerd door STS kan worden verbeterd door de expressie van functioneel caspase-3 via caspase-8-activering en Bid-splitsing. Behandeling met 1 μM remt slechts gedeeltelijk IL-3-gestimuleerde Bcl2-fosforylering, maar blokkeert volledig PKC-gemedieerde Bcl2-fosforylering. Het induceert apoptose van humane voorhuidfibroblasten AG-1518, afhankelijk van de lysosomale cathepsinen D-gemedieerde cytochroom c-afgifte en caspase-activering. Naast het activeren van de klassieke mitochondriale apoptoseroute, triggert STS een nieuwe intrinsieke apoptoseroute, die berust op de activering van caspase-9 in afwezigheid van Apaf-1.

Proteïne kinase C wordt geanalyseerd in een reactiemengsel (0,25 mL) dat 5 μmol Tris/HCl, pH 7,5, 2,5 μmol magnesiumacetaat, 50 μg histone II S, 20 μg fosfatidylserine, 0,88 μg dioleïne, 125 nmol CaCl2, 1,25 nmol [γ-32]ATP (5-10 × 104 cpm/nmol) en 5 μg gedeeltelijk gezuiverd enzym bevat. De binding van [3H]PDBu aan proteïne kinase C wordt bepaald: Reactiemengsel (200 μL) bevatte 4 μmol Tris/malaat, pH 6,8, 20 μmol KCl, 30 nmol CaCl2, 20 μg fosfatidylserine, 5 μg gedeeltelijk gezuiverde proteïne kinase C, 0,5% (eindconcentratie) DMSO, 10 pmol [3H]PDBu (1-3 × 104 cpm/pmol) en 10 μL van verschillende hoeveelheden Staurosporine (STS).
In vivo
In de gerbil- en rat-ischemiemodellen voorkomt voorbehandeling met Staurosporine (STS) (0,1–10 ng) vóór ischemie neuronale schade op een dosisafhankelijke manier, wat duidt op de betrokkenheid van PKC bij de dood van CA1-piramidecellen na ischemie.
