Beschrijving

Dit steroïde lacton, geïsoleerd uit Withania somnifera, remt krachtig de activering van NF-κB door de tumornecrosefactor-geïnduceerde activering van IκB-kinase β te voorkomen via een thioalkyleringsgevoelig redoxmechanisme. Het bindt ook aan het intermediaire filament (IF) eiwit, vimentine, met antitumor- en antiangiogenese-activiteit.

Withaferin A (WFA) remt potent NFκB-activering door de door tumornecrosefactor geïnduceerde activering van IκB-kinase β te voorkomen via een thioalkylatiegevoelig redoxmechanisme. Het bindt aan het intermediaire filament (IF) eiwit vimentine, door de cysteïneresidu ervan, aanwezig in het sterk geconserveerde α-helix spiraalvormige coiled coil 2B-domein, covalent te modificeren en induceert vimentinefilamenten om in vitro te aggregeren.

In vivo

Withaferin A (WFA) vertoont duidelijk ontstekingsremmende eigenschappen in een acuut ontstoken muismodel. De geïnduceerde remming van capillaire groei in een muismodel van corneale neovascularisatie is gecompromitteerd bij vimentine-deficiënte muizen.
