 5.000mg/ml (6.06mM) Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to make it clear; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Biologische activiteit

Beschrijving Vincristine (Leurocristine, NSC-67574, 22-Oxovincaleukoblastine) is een natuurlijk alkaloïde geïsoleerd uit de plant Vinca rosea Linn. met anti-tumoractiviteit. Deze verbinding remt de microtubulivorming in de mitotische spoel en bindt aan microtubuli met een Ki van 85 nM.
In vitro

Deze verbinding, een microtubulus-destabiliserend middel (MDA), remt de vorming van microtubuli en de mitose van de kankercel, die het slachtoffer is van intratumorale metabolische resistentie die geen belangrijke relevantie heeft met CYP3A4 noch met CYP3A5.
In vivo

Deze verbinding toont de regressie van de tumor in zebravissen-xenotransplantaten.

    HepG2 sublines (100×104 cells/well) are seeded in 12-well plates and cultured until full confluence. Then, the media is removed, and the cells are exposed to 1 μM drug (docetaxel, paclitaxel or vincristine) or 0.01% DMSO (vehicle control). Samples are collected at 24, 48 and 72 h intervals using TRI Reagent, which is followed by RNA isolation. The RNA yield is assessed via a NanoDrop ND-1000 spectrophotometer, while its quality and integrity are verified by agarose gel electrophoresis.
