MK-8931 (Verubecestat) is een krachtige en selectieve bèta-secretase (ook bekend als bèta-site APP-splitsend enzym 1) remmer.

In vitro

Verubecestat (MK-8931) vermindert effectief Aβ40 in cellen met een Ki van 7,8 nM en een IC50 van 13 nM.

In vivo

Verubecestat (MK-8931) verlaagt dramatisch CSF en cortex Aβ40 bij zowel ratten als cynomolgusapen na een enkele orale dosis. Door de halfwaardetijd van 20 uur is deze verbinding ideaal voor een eenmaal daagse dosering.
