Biologische activiteit

Beschrijving Vactosertib (TEW-7197, EW-7197) is een zeer potente, selectieve en oraal biologisch beschikbare TGF-β-receptor ALK4/ALK5-remmer met een IC50 van respectievelijk 13 nM en 11 nM. Fase 1.

In vitro In HaCaT (3TP-luc) en 4T1 (3TP-luc) stabiele cellen remt 12b potent de TGF-β1-geïnduceerde luciferase-reporteractiviteit met IC50 van respectievelijk 16,5 en 12,1 nM. EW-7197 remt TGFβ-geïnduceerde Smad2- of Smad3-fosforylering en de epitheliale-naar-mesenchymale overgang (EMT) in met TGFβ behandelde borstkankercellen. Bovendien annuleert EW-7197 ook TGFβ1-geïnduceerde tumorcelmigratie en -invasie in borstcellen.
In vivo Bij ratten vertoont EW-7197 een orale biologische beschikbaarheid van 51% met een hoge systemische blootstelling (AUC) van 1426 ng×h/mL en een maximale plasmaconcentratie (Cmax) van 1620 ng/mL. EW-7197 vertoont ook een lage toxiciteit voor het cardiovasculaire systeem, het centrale zenuwstelsel en het ademhalingssysteem. [1] In een muis B16 melanoommodel onderdrukt EW-7197 (2,5 mg/kg dagelijks p.o.) de progressie van melanoom met verbeterde cytotoxische T-lymfocyt (CTL)-responsen. EW-7197 verhoogt de cytotoxische T-lymfocytactiviteit bij 4T1 orthotoop geënte muizen en verlengde de overlevingstijd van 4T1-Luc en 4T1 borsttumor-dragende muizen.

    Cells are seeded in 96 well plate and treated with indicated concentrations of EW-7197 in 0.2% HI-FBS medium for 72 h. Cells are dried after incubation with 10% TCA in media. Then, cells are incubated with 0.4% SRB (Sulforhodamine B) in 1% acetic acid for 30 min. After washing with 1% glacial acetic acid, bounded dye is released in 10 mM Tris buffer (pH 10.5) for 30 min. Absorbance is measured at 570 nm.
