Biologische activiteit

Beschrijving Tyrphostin AG 1296 is een remmer van PDGFR met een IC50 van 0,3-0,5 μM, zonder activiteit tegen EGFR. Tyrphostin AG1296 remt FGFR en c-Kit met een IC50 van respectievelijk 12,3 μM en 1,8 μM in Swiss 3T3-cellen. Tyrphostin AG1296 induceert dramatische apoptosis in A375R-cellen.

In vitro AG 1296 remt selectief de PDGF-receptorkinase en de PDGF-afhankelijke DNA-synthese in Swiss 3T3-cellen en in endotheelcellen van de varkensaorta met 50% remmende concentraties onder respectievelijk 5 en 1 μM. AG1296 remt FGFR en c-Kit met een IC50 van 12,3 μM en 1,8 μM in Swiss 3T3-cellen. AG1296 remt potent de signalering van humane PDGF-α- en -β-receptoren, maar heeft geen effect op de autofosforylering van de VEGFR KDR of op de door VEGF geïnduceerde DNA-synthese in endotheelcellen van de varkensaorta. Behandeling met AG1296 keert het getransformeerde fenotype van sis-getransfecteerde NIH 3T3-cellen om, maar heeft geen effect op src-getransformeerde NIH3T3-cellen. AG1296 is een ATP-competitieve remmer. AG1296 interfereert noch met PDGF-binding noch met PDGF-receptordimerisatie, terwijl het de PDGF-receptorautofosforylering opheft. Aldus is AG1296 een zuivere remmer van de katalytische activiteit van de receptor-tyrosinekinase.

  • Membraan-autofosforyleringstesten

    Membranen worden bereid uit concurrerende culturen van Swiss 3T3-cellen zoals beschreven. Voor het meten van receptor-autofosforylering wordt 10 μg membraanproteïne per test gedurende 20 min op ijs geïncubeerd in aanwezigheid van 1,2 μg/mL EGF of 2 μg/mL PDGF, of beide; 50 mM Hepes (pH 7,5); en 3 mM MnCl2 in een volume van 45 μl. Om de effecten van tyrfostines te testen, worden deze toegevoegd in een volume van 0,5 μl (in DMSO; eindconcentratie, 0,5%) 15 min vóór toevoeging van de groeifactoren. Fosforylering wordt geïnitieerd door toevoeging van [γ-32P]ATP en beëindigd na 2 min door toevoeging van 10 μL van een oplossing die 6% SDS, 30% β-mercaptoethanol, 40% glycerol en 0,5 mg/mL broomfenolblauw bevat. De monsters worden gedurende 5 min verwarmd op 95 ℃ en onderworpen aan SDS-PAGE met 10% acrylamidegels. De gels worden gekleurd en gedroogd en onderworpen aan autoradiografische analyse.

<p>After inoculation with KAT4 cells, administration of compounds including MK-2206 (100 mg/kg), tyrphostin AG 1296 (100 mg/kg), or a combination of MK-2206 (100 mg/kg) and tyrphostin AG 1296 (100 mg/kg) was performed. The combination of MK-2206 and tyrphostin AG 1296 induced significant apoptosis of KAT4 tumor cells in vivo measured by the terminal deoxynucleotidyl transferase dUTP nick end labeling assay (green), and nuclei were stained with Hoechst (blue).</p>
