Biologische activiteit

Beschrijving Tocilizumab (anti-IL-6R) is een gehumaniseerd monoklonaal antilichaam dat bindt aan de interleukine-6 receptor, MW: 148 KD.

In vitro

Na toevoeging van tocilizumab in verschillende concentraties aan HMO6- en CHME5-cellen gestimuleerd door F3-CM of met IL6, nemen de expressieniveaus van ontstekingsremmende M2-markers (CD206 en Arg1) af, evenals de niveaus van gefosforyleerde JAK2 en STAT3.
In vivo

Tocilizumab kan JAK2/STAT3-activatie blokkeren en hersenmetastase (BM) van A549-F3-cellen in vivo tegengaan.

Protocol (alleen ter referentie)

Celassay:

  • HMO6- en CHME5-cellen met controlemedia of F3-CM alleen of F3-CM werden behandeld met Tocilizumab (2,5 μg/ml gedurende 24 h) of met IL6 (200 ng/ml gedurende 24 h) alleen of IL6-aangevuld met Tocilizumab. Western blot-analyse van p-JAK2, JAK2, p-STAT3 en STAT3 in HMO6- en CHME5-cellen werd uitgevoerd.
Dierstudie:

  • Dierlijke modellen: Naakte muizen met hersenmetastasemodellen
