In vitro

In NIH 3T3-cellen remt SU 6656 de PDGF-gestimuleerde S-fase-inductie met een IC50 van 0,3-0,4 μM. SU 6656 remt ook PDGF- en serum-gemedieerde NIH 3T3-celproliferatie, evenals epidermale groeifactor en colony-stimulating factor 1-gestimuleerde DNA-synthese in normale en colony-stimulating factor 1-receptor-getransfecteerde NIH 3T3-cellen. SU6656 remt PDGF-gestimuleerde c-Myc-inductie en ERK2-activering. Voorbehandeling van Jurkat T-cellen met SU 6656 leidt tot verhoogde VSV-G luciferase-activiteit. SU 6656 vermindert TGF-β-gemedieerde opregulatie van CTGF mRNA en eiwit in proximale epitheliale HKC-8-cellen, en vermindert ook de CTGF-expressie in cellen die zijn blootgesteld aan autocriene groeifactoren. SU 6656 interfereert met Aurora kinase-activiteit, wat resulteert in remming van celdeling en de vorming van multilobulaire kernen.
In vivo

SU 6656 verzwakt de -geïnduceerde experimentele nicotine-ontwenningssyndroom bij muizen, gemeten in termen van WSS en angstsymptomen, aanzienlijk en dosisafhankelijk.

  • Biochemische kinasetesten voor IC50-bepaling en kinetische studies

    IC50-metingen worden uitgevoerd met poly-Glu-Tyr (4:1), of, in het geval van Lck, poly-Lys-Tyr (4:1) als peptidesubstraat. Het tweewaardige kation is 20 mM MgCl2 (in het geval van Src, Fyn, Yes, Lyn, Csk, Frk of Abl) of 10 mM MnCl2 (in het geval van FGFR1, IGF1R, Lck of Met).
