Beschrijving STM2457 is een zeer potente en selectieve 'first-in-class' katalytische remmer van RNA Methyltransferase METTL3 met een IC50 van 16,9 nM. STM2457 is zeer specifiek voor METTL3 en vertoonde geen remming van andere RNA methyltransferases.

Deze verbinding heeft een zeer efficiënte en selectieve katalytische inhibitie op RNA methyltransferase METTL3, wat genexpressiedefecten veroorzaakt die consistent zijn met een effect op de mRNA translationele efficiëntie van m6A substraten.
In vivo

STM2457 voorkomt de expansie van acute myeloïde leukemie (AML) en vermindert het aantal belangrijke leukemie stamcellen in vivo.
