 2.100mg/ml (9.94mM) Taking the 1 mL working solution as an example, add 50 μL of 42 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Biologische activiteit

Beschrijving Stattic, het eerste niet-peptidische kleine molecuul, remt krachtig de STAT3-activering en nucleaire translocatie met een IC50 van 5,1 μM in celvrije assays, met hoge selectiviteit ten opzichte van STAT1. Stattic induceert apoptose.

In vitro

Stattic remt de binding van een fosfotyrosine-bevattend peptide afkomstig van de gp130-receptor aan het STAT3 SH2-domein op een sterk temperatuurafhankelijke manier. Deze verbinding heeft slechts een zeer zwak effect op de binding van een tyrosine-gefosforyleerd peptide aan het SH2-domein van de tyrosinekinase Lck. En het remt de dimerisatie van twee andere dimere transcriptiefactoren (c-Myc/Max en Jun/Jun) niet. Het remt ook fluoresceïne-gelabelde fosfopeptiden aan de SH2-domeinen van STAT1 en STAT5b. Deze chemische stof remt selectief de DNA-binding van STAT3-homodimeren bij een concentratie van 10 μM. Het is aangetoond dat het cellulaire fosforylering van STAT3 op Tyr705 remt met weinig effect op STAT1-fosforylering op Tyr701 (in HepG2-cellen) of de fosforylering van JAK1, JAK2 en c-Src (in MDA-MB-231- en MDA-MB-235S-cellen). Deze verbinding verhoogt de apoptotische snelheid van STAT3-afhankelijke borstkankercellijnen.

Kenmerken Stattic is het eerste niet-peptide kleine molecuul met remmende activiteit tegen het STAT3 SH2-domein, ongeacht de STAT3-fosforylatietoestand in vitro.

  • High-Throughput Screening en fluorescentiepolarisatie-assays

    Screening wordt uitgevoerd bij ongeveer 30 °C. De specificiteit van de screeninghits wordt gevalideerd in analoge assays voor de binding van de testverbindingen aan de SH2-domeinen van STAT1, STAT5 en Lck. De uiteindelijke concentratie van buffercomponenten die voor alle FP-assays wordt gebruikt, is 10 mM HEPES (pH 7,5), 1 mM EDTA, 0,1% Nonidet P-40, 50 mM NaCl en 10% DMSO. De afwezigheid van dithiothreitol is essentieel voor de remmende activiteit. De sequenties van de peptiden zijn: STAT3, 5-carboxyfluoresceïne-GY(PO3H2)LPQTV-NH2; STAT1, 5-carboxyfluoresceïne-GY(PO3H2)DKPHVL; STAT5, 5-carboxyfluoresceïne-GY(PO3H2)LVLDKW; en Lck, 5-carboxyfluoresceïne-GY(PO3H2)EEIP. Voor specificiteitsanalyse bij 30 °C worden eiwitten gebruikt bij 150 nM (STAT1, STAT3 en STAT5). Voor specificiteitsanalyse bij 37 °C worden eiwitten gebruikt bij 370 nM (STAT3) of 100 nM (Lck). Eiwitten worden in Eppendorf-buisjes met testverbindingen geïncubeerd bij de aangegeven temperaturen gedurende 60 minuten voorafgaand aan de toevoeging van de respectieve 5-carboxyfluoresceïne-gelabelde peptiden (laatste concentratie: 10 nM). Vóór meting bij kamertemperatuur laat men de mengsels ten minste 30 minuten in evenwicht komen. Testverbindingen worden gebruikt in de aangegeven concentraties, verdund uit een 20× stamoplossing in DMSO. Bindingscurven en remmingscurven worden gefit met SigmaPlot. Alle competitiecurven worden driemaal herhaald in onafhankelijke experimenten.
