Biologische activiteit

Beschrijving Spautin-1 is een potente en specifieke autophagy-remmer en remt de deubiquitinerende activiteit van USP10 en USP13 met een IC50 van ~0,6-0,7 μM. Deze verbinding versterkt apoptosis.

In vitro

Spautin-1 bevordert de afbraak van Vps34-complexen door de deubiquitinatie-activiteit van USP10 en USP13 te reguleren, en verlaagt de niveaus van PtdIns3P. In met PDGF behandelde cellen stabiliseert deze verbinding α-gladde spiercelactine en calponine, voorkomt desorganisatie van actinefilamenten, vermindert de productie van extracellulaire matrix en heft VSMC-hyperproliferatie en -migratie op. In CML-cellen remt het de IM-geïnduceerde autofagie aanzienlijk door Beclin-1 down te reguleren, en verhoogt het de IM-geïnduceerde apoptose door PI3K/AKT te inactiveren en stroomafwaartse GSK3β te activeren. Deze chemische stof vermindert ook specifiek infectieuze denguevirus-titers in BHK-21-cellen.

Autophagosomal inhibition was performed in 32D EV, WT CALR, del52, and ins5 expressing cells. The 32D cell lines were treated with the inhibitor spautin-1 for the indicated time. Lysates were prepared, and SDS-PAGE and Western blotting were performed. Antibodies detecting mutated CALR (CALR mut), CALR, LC3I-II, and GAPDH were used for immunostaining. LC3I-II served as control for successful autophagosomal inhibition.
