Selonsertib (GS-4997) is een zeer selectieve en potente eenmaal daagse orale ASK1-remmer met een pIC50 van 8,3.

In vivo

Een oraal biologisch beschikbare remmer van apoptosis signal-regulating kinase 1 (ASK1), met potentiële ontstekingsremmende, antineoplastische en antifibrotische activiteiten. Na orale toediening richt de ASK1-remmer GS-4997 zich op en bindt zich aan het katalytische kinasedomein van ASK1 op een ATP-competitieve manier, waardoor de fosforylering en activering ervan wordt voorkomen. Dit voorkomt de fosforylering van stroomafwaartse kinasen, zoals c-Jun N-terminale kinasen (JNK's) en p38 mitogeen-geactiveerd proteïnekinase (p38 MAPK). Door de activering van ASK1-afhankelijke signaaltransductiepaden te voorkomen, voorkomt GS-4997 de productie van ontstekingscytokines, reguleert het de expressie van genen die betrokken zijn bij fibrose neerwaarts, onderdrukt het overmatige Apoptosis en remt het de cellulaire proliferatie.
