Beschrijving SC75741 is een potente NF-κB-remmer met een EC50 van 200 nM. Deze verbinding blokkeert efficiënt de verspreiding van het Influenza Virus.

In vitro SC75741 vertoont immuunsuppressieve activiteit door de proliferatie van humane PBMC's te remmen met een IC50 van 2,2 μM. Deze verbinding remt de replicatie van influenza A- en B-virussen door NF-κB-gemedieerde signalering op transcriptioneel niveau te remmen. Bovendien toont het een hoge barrière voor de ontwikkeling van resistente virusvarianten.
In vivo SC75741 (15 mg/kg i.p.) vermindert virusreplicatie en cytokine-expressie in muizenlongen na H5N1 Influenza Virus-infectie.

  • NF-κB reportergen assay

    De NF-κB reportergen assay wordt voorbereid met de A549-NF-κB-SEAP-cellijn volgens de instructies van de fabrikant. Kortom, A549-cellen die stabiel getransfecteerd zijn met het pNF-κB-SEAP reportergen-plasmide worden geënt en overnacht gehecht. De cellen worden vervolgens 5 uur geïncubeerd met beschreven verbindingen bij 100, 30, 10, 3, 1, 0,3, 0,1 en 0 μM en daarna gedurende 22 uur gestimuleerd met 10 ng/ml TNF-α. Het supernatant van de cel wordt geanalyseerd op SEAP-activiteit met behulp van een chemiluminescentie SEAP reportergen assay.

SK-OV-3 and A2780S cells were treated with different compounds (vehicle, 5 μM JSH-23, 5 μM SC75741, 5 μM CAPE, 4 μM MIL, 4 μM MIL combined with pre-treated 5 μM JSH-23, 4 μM MIL combined with pre-treated 5 μM SC75741, 4 μM MIL combined with pre-treated 5 μM CAPE) for 48 h.
