Biologische activiteit

Beschrijving Entecavir (BMS200475, SQ34676), een nieuw deoxyguaninenucleoside-analogon, is een selectieve remmer van de replicatie van het hepatitis B-virus (HBV).
In vitro

Voor de behandeling van hepatitis B-virus leiden combinaties van ETV met elk van de andere HBV NRTI-antivirale middelen, adefovir, tenofovir en lamivudine, bij 10 keer hun respectieve Cmax-niveaus niet tot cellulaire of mitochondriale toxiciteit en vertonen ze geen DNA-polymerase gamma-inhibitie.
In vivo

Behandeling met Entecavir in combinatie met AIC649 induceert strak gereguleerde immunologische en virologische responsen tijdens actieve en onderhoudsbehandeling in het marmotmodel van chronische hepatitis B.

    HepG2 cells (2.7 × 106) are seeded onto collagen I-coated T75 flasks. Cultures are exposed to compounds for a total of 15 days, reseeding every 5 days to the original cell density. In addition, media and compounds are also replaced 3 days after seeding. This schedule results in replacement of media containing fresh compound on days 1, 3, 5, 8, 10, and 13. Cells and culture supernatants are harvested and assayed on days 5, 10, and 15.
