 4.950mg/ml (9.91mM) Taking the 1 mL working solution as an example, add 50 μL of 99 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Biologische activiteit

Beschrijving (S,S)-TAPI-1 is een isomeer van TAPI-1. TAPI-1 is een ADAM17/TACE-remmer, die het afstoten van cytokinereceptoren blokkeert. TAPI-1 is ook een remmer van Matrix Metalloproteinase (MMP).

In vitro

TAPI-1 voorkomt ongestimuleerde en PMA-geïnduceerde afgifte van de oplosbare vormen van TNF-alfa, p60 TNFR en IL-6R uit de monocytaire cellijn THP-1 en uit menselijke perifere bloedmonocyten. TAPI remt ook LPS-geïnduceerde afscheiding van de p60 TNFR en TNF-alfa uit monocyten.

TAPI-1 remt TACE-afhankelijke constitutieve afgifte van co-getransfecteerd APP(695).

TAPI-1 vermindert Ang II-geïnduceerde EGFR-transactivatie en celproliferatie in menselijke HSC-lijn LI90.

TAPI-1 met de EGFR-remmer AG1478 vertoont een gedeactiveerde AREG/EGFR/ERK-signaleringsroute en vermindert de afgifte van pro-inflammatoire cytokines in pSS-speekselklier-afgeleide epitheelcellen.

    Five thousand cells are seeded into each well of 96-well plates and their viability is assessed by CellTiter-Glo Luminescent Cell Viability assay. Upon serum deprivation for 24 h, the cells are treated with Ang II for 60 h with and without pretreatment with each inhibitor and antagonist. The assay substrates are then added to each well on the plate and the samples are evaluated using a luminometer.
