Beschrijving RU.521 (RU320521) is een actieve, potente en selectieve remmer van cyclic GMP-AMP synthase (cGAS) die de cGAS-gemedieerde signalering remt met een IC50 van 700 nM. RU.521 bindt aan het cGAS/dsDNA-complex met een Kd van 36,2 nM.

In vitro

RU.521, een cGAS-remmer, vermindert de overexpressie van interferon-gestimuleerde genen (ISG's) als reactie op cytolethaal distenderend toxine (CDT), wat de cruciale rol van de cGAS-STING-as in CDT-geïnduceerde type I IFN-signalering bevestigt.
In vivo

RU.521 verlichtte de ontstekingsreactie en verminderde de schade veroorzaakt door oxidatieve stress, wat leidde tot cardiale bescherming via verhoogde Sirt3-expressie in de harten van muizen met sepsis.

  • Methode

    MEFs are exposed with or without STINGi or cGASi to this compound 7.5 ng/ml for the indicated times or to the H153A CDT mutant for 40 days. The mRNA level of the indicated genes is analyzed by RT-qPCR and the IL-6 or TNFα production in the culture supernatant are determined by ELISA. For ELISA assays, a 2 days exposure to LPS 100 ng/ml is used as positive control.
