 0.300mg/ml (0.96mM) Taking the 1 mL working solution as an example, add 50 μL of 6 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Beschrijving RIN1 (RBPJ Inhibitor-1) is een krachtige remmer van de transcriptiefactor RBPJ die de interactie tussen NOTCH en RBPJ verstoort.

RBPJ Inhibitor-1 (RIN1) blokkeert de functionele interactie van RBPJ met SHARP, een scaffold-eiwit dat een transcriptioneel repressorcomplex vormt met RBPJ in afwezigheid van NOTCH-signalering. In overeenstemming met de verstoring van NOTCH-signalering remt deze verbinding de proliferatie van hematologische kankercellijnen en bevordert het de differentiatie van skeletspieren uit C2C12-myoblasten.

    AD-293 cells were transfected with RBPJ-VP16myc and 48 hours later were treated±RIN1 (RBPJ Inhibitor-1, 2 µM) and Cycloheximide (CHX, 10 µg/ml) for an additional 17 hours, then assayed for Hes1-Luciferase activity.
