Biologische activiteit

Beschrijving Purvalanol A is een potente en celpermeabele CDK-remmer met een IC50 van respectievelijk 4 nM, 70 nM, 35 nM en 850 nM voor cdc2-cycline B, cdk2-cycline A, cdk2-cycline E en cdk4-cycline D1. Deze verbinding induceert endoplasmatisch reticulumstress-gemedieerde apoptosis en autophagy.

In vitro Purvalanol A vermindert de cellevensvatbaarheid dosisafhankelijk in MCF-7- en MDA-MB-231-cellijnen. Deze verbinding induceert 50% verlies van cellevensvatbaarheid in MCF-7-cellen, maar MDA-MB-231-cellen zijn er minder gevoelig voor (32% afname in cellevensvatbaarheid). Het induceert mitochondriën-gemedieerde apoptosis in MCF-7- en MDA-MB-231-cellen. Deze chemische stof voorkomt effectief c-Src-gemedieerde transformatie door zowel de Cell Cycle-progressie als de c-Src-signalering te remmen, en onderdrukt effectief de verankering-onafhankelijke groei van sommige menselijke kankercellen waarin c-Src is opgereguleerd. Het heeft een sterker remmend effect op de verankering-onafhankelijke groei van HT29- en SW480-menselijke darmkankercellen.

    Cells are seeded at 10000 density in 96-well plates and treated with various concentrations of Purvalanol A (0-100 μM) for 24 h. They are exposed to 10 μL of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltet- razolium bromide dye (5 mg/mL) and are incubated at 37℃ for 4 h. In order to solubilize the formazan crystals 100 μL DMSO is added. Absorbance is determined at 570 nm spectrophotometrically.
