Biologische activiteit

Beschrijving Necrosulfonamide is een zeer specifieke en potente necrose-remmer en blokkeert mixed lineage kinase domain-like protein (MLKL).

In vitro

Necrosulfonamide remt MLKL-gemedieerde necrose door de functie van het N-terminale CC-domein te blokkeren. Het blokkeert necrose stroomafwaarts van RIP3-activering. Deze verbinding heeft geen effect op apoptose geïnduceerd door TNF-α plus Smac-mimeticum in niet-RIP3-expressende Panc-1-cellen, zelfs niet bij een concentratie van 5 μM. De reden voor de soortspecificiteit van deze chemische stof is dat de cysteïne op residu 86 in humaan MLKL dat covalent gemodificeerd wordt door de stof, wordt vervangen door een tryptofaanresidu in muis MLKL (mixed lineage kinase domain-like protein).
In vivo

Necrosulfonamide (NSA) is een klein molecuul dat necroptose specifiek remt door zich te richten op MLKL, de terminale uitvoerder van dit proces.

    Necrosis inhibitors induce diverse effects on MLKL phosphorylation. HT-29 cells are treated with T/S/Z with or without necrosis inhibitors for 12 hr or 8 hr. The number of dead cells is determined by measuring released protease activity in culture medium. The whole-cell extracts are prepared and analyzed by western blotting. The final concentrations of 10 μM necrostatin-1 or 1 μM this compound are used to block necrosis.

Necroptotic death of HT-29 cells was verified using RIP1 or MLKL inhibitors necrostatine-1 and necrosulfonamide. HT-29 were pre-treated with the combinations of 10 nM birinapant, 50 μM z-VAD, 50 μM necrostatin and 20 μM necrosulfonamide as shown for 1 h, and then treated with 100 ng/ml of TST-TRAIL-wt, 4.2, 5.1 and 5.2 for 8 h.
