  • Kinase Assays

    Initiële ULK1 kinase assays worden uitgevoerd met GST-ULK1, geproduceerd in Sf9-cellen, zoals beschreven op de MRC-PPU Reagents website. Voor andere experimenten wordt recombinant GST-ULK1 (wild type, kinase-dood (K46I), en M92T en M92Q) tot expressie gebracht in 293T-cellen, gezuiverd en geëlueerd van een glutathion-Sepharose kolom. Kinase assays worden uitgevoerd in 50 mM Tris-HCl, pH 7.4, 10 mM magnesiumacetaat, 0,1 mM EGTA en 0,1% β-mercaptoethanol, met 30 μM koud ATP en 0,5 μCi [γ-32P]ATP gedurende 5 min bij 25 °C. Voorafgaand aan de ATP-toevoeging worden reactiemengsels gedurende 5 min voorverwarmd tot 25 °C. Reacties worden gestopt door toevoeging van samplebuffer, gevolgd door SDS-PAGE, overdracht naar nitrocellulose en analyse door autoradiografie en immunoblot.

    MEFs and 293T cells were grown in DMEM, supplemented with 10% fetal bovine serum and penicillin/streptomycin, and cultured at 37 °C, 5% CO2. For induction of autophagy, cells were typically grown to 75% confluency, washed twice, and incubated in Earle's balanced salt solution (EBSS) for 1 h (or complete medium as a control) in the presence or absence of 10 μM MRT67307, 1 μM MRT68921, or 50 nM bafilomycin A1 and followed by lysis and immunoblotting with the indicated antibodies.

In 3D multicellular spheroids with high autophagy, MRT 68921 interferes with autophagy initiation. MCS with either low (REN - low ATG) or high autophagy (M28-high ATG) at steady state were exposed to MRT, HCQ or the vehicle control (-) as in Fig.1. Spheroids were disaggregated and the cells cytospun on glass slides. Cells were fixed, stained for ATG13 (green) and for nuclei (TO-PRO-3, blue), and imaged by confocal microscopy. Representative ATG13 puncta are indicated by arrows. In REN-low ATG, no ATG13 puncta were seen, as we have shown previously,35 indicating that these cells have inactive initiation. As expected, in REN, neither MRT nor HCQ had any effect. In M28-high ATG, ATG13 puncta were prominent at steady state, as we have shown previously,35 indicating that these cells have active initiation. In these cells, after MRT, ATG13 puncta were no longer detectable, confirming that MRT interfered with the initiation of autophagy. In M28, HCQ had no effect on initiation. Scale bars: 10 μm.
