MRT67307 is een potente en dubbele IKKϵ- en TBK1-remmer met een IC50 van respectievelijk 160 en 19 nM. MRT67307 remt krachtig ULK1 en ULK2 en blokkeert autophagy.

In vitro

In macrofagen voorkomt MRT67307 de fosforylering van IRF3 en de productie van IFNβ. In wildtype MEF's versterkt MRT67307 de IL-1-gestimuleerde fosforylering van p105 bij Ser933 en RelA bij zowel Ser468 als Ser536, en versterkt het ook de IL-1-gestimuleerde activering van NF-κB-afhankelijke gen transcriptie. MRT67307 verhoogt de IL-10-productie en onderdrukt de productie van pro-inflammatoire cytokines via een cAMP response element-Binding protein (CREB)-gereguleerd transcriptioneel coactivator (CRTC) 3-afhankelijk mechanisme. Bovendien remt MRT67307 ULK en blokkeert het autophagy in MEF-cellen.
