Motolimod is een selectieve en potente Toll-like receptor 8 (TLR8) agonist met een EC50 van 100 nM, > 50-voudige selectiviteit ten opzichte van TLR7. Fase 2.

In vitro
VTX-2337 stimuleert de productie van zowel TNFα met een EC50 van 140 nM als IL-12 met een EC50 van 120 nM in PBMC's. In monocyten en mDC's induceert VTX-2337 selectief de productie van TNFα en IL-12 via NF-κB activering. VTX-2337 stimuleert ook IFNγ-productie door NK-cellen, verhoogt de lytische functie van NK-cellen en verbetert ADCC.
In vivo
In een muismodel voor eierstokkanker versterkt TX-2337 het effect van gepegyleerde liposomale doxorubicine (PLD).
De activiteit van specifieke TLR-agonisten wordt beoordeeld met behulp van het secretoir embryonaal alkalisch fosfatase (SEAP) reportergen dat is gekoppeld aan NF-κB-activering als reactie op TLR-stimulatie. Meting van SEAP-activiteit met behulp van het Quanti-blue substraat (InvivoGen) na TLR-agonistbehandeling wordt uitgevoerd.
PBMCs or purified NK cells are prepared as previously described, and the purity of NK cells was approximately 99%. NK cell–mediated cytotoxicity is assessed by Calcein AM release from labeled target cells. In brief, PBMCs or purified NK cells are cultured for 48 hours in RPMI medium in the presence of VTX-2337 (167 or 500 nmol/L) before incubation with target cells.
