Beschrijving

ML-792 is een potente en selectieve remmer van het SUMO (small ubiquitin-like modifier)-activerende enzym (SAE). ML-792 remt SAE/SUMO1 en SAE/SUMO2 in ATP-anorganisch pyrofosfaat (PPi) uitwisselingstests met IC50 van respectievelijk 3 nM en 11 nM.

In vitro

ML-792 is een mechanisme-gebaseerde SUMO-activerende enzym (SAE) remmer met nanomolaire potentie in cellulaire assays. ML-792 blokkeert selectief de SAE enzymactiviteit en totale SUMOylering, waardoor de proliferatie van kankercellen afneemt. Inductie van het MYC-oncogen verhoogt het ML-792-gemedieerde levensvatbaarheidseffect in kankercellen, wat wijst op een potentiële toepassing van SAE-remmers bij de behandeling van MYC-geamplificeerde tumoren.
