Beschrijving ML323 vertoont reversibele, nanomolaire remmende activiteit en uitstekende selectiviteit ten opzichte van USP1/UAF1 met een IC50 van 76 nM.

In vitro

ML323 remt de deubiquitinatie van PCNA en FANCD2 door de USP1-UAF1-activiteit in H596-cellen te remmen. Bovendien versterkt deze verbinding de cytotoxiciteit in NSCLC H596-cellen en U2OS-osteosarcoomcellen door twee belangrijke DNA-schaderesponspaden (TLS en FA) aan te pakken.

In vivo

Deze verbinding is een selectieve USP1-UAF1-remmer die de deubiquitinase-activiteit van het USP1-UAF1-complex remt.

  • High-throughput screening

    Voor HTS wordt de USP1-UAF1-activiteit gemonitord met behulp van Ubiquitin-rhodamine 110 als substraat. De assay is geminiaturiseerd tot een volume van 4 μL in een 1.536-well formaat en wordt gebruikt om ongeveer 402.701 verbindingen te screenen in kwantitatieve HTS-modus, waarbij elke verbinding wordt getest over een bereik van vier tot vijf concentraties. De assay toont robuuste prestaties met een gemiddelde Z'-factor van 0,8 gedurende het hele scherm.

    For the colony-forming assay, cells are seeded at a density of 300–500 cells per well in six-well plates and grown overnight. Cells are then treated with ML323 alone at the indicated concentrations. Cells treated with an equal volume of DMSO and saline are used as control. After 48 h of treatment, fresh growth medium is added, and cells are incubated for an additional 5-10 d to allow for colony formation. For UV combination treatment, the cells are treated with this compound at the indicated concentrations or an equal volume of DMSO. After 48 h, the medium is removed, and cells are irradiated at 254 nm at the indicated dosage. Fresh growth medium is added, and the cells are incubated for an additional 5-10 d to allow for colony formation. The cells without UV irradiation but treated with this chemical or an equal volume of DMSO are used as controls and designated as 100%. After the formation of the colonies, cells are fixed with methanol and stained with 0.5% crystal violet. Colonies consisting of >50 cells are scored. The number of colonies is determined from triplicate plates. The dose-response curves are generated using GraphPad Prism and analyzed by using CalcuSyn to calculate the combination index.
