Beschrijving SAR405838 is een oraal beschikbare MDM2-antagonist met een Ki van 0,88 nM. Deze verbinding is een diastereomeer van MI-773 (CAS: 1303607-07-9). Fase 1.

In vitro MI-773 bindt aan MDM2 met een Ki van 0,88 nM. Deze verbinding remt de celgroei krachtig in kankercellijnen, waaronder SJSA-1 (IC50, 0,092 μM), RS4;11 (IC50, 0,089 μM), LNCaP (IC50, 0,27 μM) en HCT-116 (IC50, 0,20 μM) cellen, en vertoont een hoge selectiviteit ten opzichte van kankercellijnen met gemuteerd of gedeleteerd p53, waaronder SAOS-2 (IC50, >10 μM), PC-3 (IC50, >10 μM), SW620 (IC50, >10 μM) en HCT-116 (p53-/-) (IC50, >20 μM) cellen.
In vivo In het SJSA-1 osteosarcoom, acute lymfatische leukemie RS4;11, LNCaP prostaatkanker en HCT-116 darmkanker xenograftmodel, remt deze verbinding (p.o.) de tumorgroei effectief op een dosisafhankelijke manier (10 mg/kg, 30 mg/kg, 50 mg/kg, 100 mg/kg en 200 mg/kg).
