Biologische activiteit

Beschrijving MCC950 Sodium is een potente, selectieve remmer van NLRP3 met een IC50 van 7,5 nM in BMDM's; maar niet van de AIM2, NLRC4 of NLRP1 inflammasomen.

In vitro

MCC950 remt canonieke en niet-canonieke NLRP3-activering in BMDM- en HMDM-cellen met een IC50 van 7,5 en 8,1 nM. MCC950 remt specifiek de activering van NLRP3, maar niet die van de AIM2-, NLRC4- of NLRP1-inflammasomen.
In vivo

MCC950 vermindert de interleukine-1β (IL-1β)-productie en vermindert de ernst van experimentele auto-immune encefalomyelitis (EAE). Bovendien redt de MCC950-behandeling neonatale letaliteit in een muismodel van CAPS en is het actief in ex vivo-monsters van individuen met het Muckle-Wells-syndroom.
