Biologische activiteit

Beschrijving LB-100 is een wateroplosbare proteïnefosfatase 2A (PP2A) remmer met IC50's van 0,85 M en 3,87 M in BxPc-3 en Panc-1 cellen.
Doelen
PP2A
In vitro LB-100 remt de celgroei met een IC50 van 2,3 M (in BxPc-3) of 1,7 M (in Panc-1 cellen). In BxPc-3, Panc-1 en SW1990 cellen vermindert deze verbinding de PP2A-activiteit met 30-50%. Het verhoogt de concentratie doxorubicine in cellen (2,5 keer ten opzichte van de controle) en maakt tumorcellen gevoelig voor de cytotoxiciteit van doxorubicine. Deze chemische stof verhoogt de VEGF-secretie en verbetert zo de HIF-1 -VEGF-gemedieerde angiogenese.
In vivo In een muis pancreaskanker xenograftmodel verbetert LB-100 (2 mg/kg, i.p.) de chemotherapie met doxorubicine. Deze verbinding veroorzaakt een hogere dichtheid van microvaten in tumoren en een snelle bloedstroom aan het oppervlak van tumoren.

  • PP2A-activiteitsassays

    Gekweekte pancreaskankercellen worden gedurende 2 uur behandeld met de IC50 van LB-100 voor elke cellijn of een gelijk volume van het vehikel, waarna PP2A-activiteitsassays worden uitgevoerd met behulp van een Ser/Thr-fosfataseassaykit. Cellen worden gelyseerd met een ultrasone celdisruptor en de PP2A-concentratie wordt gemeten met behulp van een Ser/Thr-fosfataseassaykit volgens de instructies. Assays voor elke cellijn worden in drievoud uitgevoerd.
