Biologische activiteit

Beschrijving	L-Kynurenine ((S)-Kynurenine) is een aryl hydrocarbon receptor agonist. Deze verbinding is een metaboliet van het aminozuur L-tryptofaan dat wordt gebruikt bij de productie van niacine en een centrale verbinding van de tryptophan Metabolism pathway.
In vitro	L-Kynurenine (Kyn), de tryptofaan (Trp) metaboliet en een AhR-ligand in hepatocyten, hoopt zich op tot cellulaire niveaus om een verhoogde snelheid van AhR-activering te veroorzaken, en stimuleert snel de transcriptionele activiteit van AhR. Een overmatige productie van deze verbinding via verhoogde cellulaire IDO1-niveaus en -activiteit leidt tot versterkte AhR-signalering die uiteindelijk resulteert in obesitas.
In vivo	L-kynurenine vertoont een dosisafhankelijk significant neuroprotectief effect vóór de hypoxie-ischemie bij ratten, en blokkeert de inductie van c-fos immunoreactiviteit in de hersenschors na ischemie.

After the 24-hr incubation, the medium is removed, and the cells are washed once with Dulbecco's Phosphate Buffered Saline. Cells are treated with this compound for an additional 24 hr, all of which are dissolved in DMSO unless otherwise noted. Then, cells are removed from incubation and allowed to equilibrate to room temperature for 15min. After equilibration, the medium is removed and the cells are washed twice with at room temperature with DPBS. The cells are lysed with 20 μl/well 1 Passive Lysis Buffer and shaken for 20 min at room temperature. Luciferase activity is recorded programmed to inject 50 μl Luciferase Assay Reagent per well with a 10 sec integration of emitted luminescence.
