Beschrijving KU-0060648 is een dubbele remmer van DNA-PK en PI3Kα, PI3Kβ, PI3Kδ met een IC50 van respectievelijk 8,6 nM en 4 nM, 0,5 nM, 0,1 nM, en een lagere remming van PI3Kγ met een IC50 van 0,59 μM.

In vitro

KU-0060648 vertoont differentiële effecten op groeiremming, maar is niet diepgaand cytotoxisch in een panel van menselijke kankercellijnen. Het remt DNA-PK en PI-3K met grotere potentie in MCF7- dan in SW620-cellen met behulp van celgebaseerde assays. Blootstelling van vijf dagen aan 1 mM van deze verbinding remt de celproliferatie met meer dan 95% in MCF7-cellen, maar slechts met 55% in SW620-cellen.
