Beschrijving GI254023X (GI 4023, SRI028594) is een selectieve remmer van ADAM10 met een 100-voudige selectiviteit voor ADAM10 boven ADAM17. De IC50-waarden voor recombinant ADAM10 en ADAM17 zijn respectievelijk 5,3 nM en 541 nM. GI254023X kan MMP9 remmen.

In celgebaseerde klievingsexperimenten blokkeert GI254023X potent de constitutieve afgifte van IL6R, CX3CL1 en CXCL16, wat in lijn is met de gerapporteerde betrokkenheid van ADAM10 maar niet ADAM17 in dit proces. Dit compound heeft daarentegen geen invloed op de PMA-geïnduceerde shedding. Naast ADAM10 en ADAM17 is deze chemische stof een matige remmer van ADAM9 met een IC50 van 280 nM.
In vivo

Acute behandeling met GI254023X in een AD-muismodel vermindert de afbraak van LRP1 in de hersenen aanzienlijk en verhoogt de Aβ40-spiegels in het plasma, wat wijst op een verbeterd Aβ-transport van de hersenen naar de periferie. Bovendien zijn zowel oplosbare als onoplosbare Aβ40- en Aβ42-niveaus in de hersenen verlaagd na behandeling met dit compound, maar deze effecten missen statistische significantie.

    HCA-7 cells are treated for 24 h with 0.1% DMSO, the metalloprotease inhibitors GI (GI254023X, ADAM10-selective, 3 µM) and GW (GW280264X, ADAM10- and ADAM17-selective, 3 µM), the pan-metalloprotease inhibitor Marimastat (MM; 10 µM), and the EGF-R kinase inhibitor AG1478 (10 µM). Cells stimulated with 100 ng/ml EGF served as positive control. Tyrosine-phosphorylated EGF-R and total EGF-R are assessed by Western blot analysis.
