FK866 (Daporinad) remt effectief nicotinamide fosforibosyltransferase (NMPRTase; NAMPT) met een IC50 van 0,09 nM in een celvrije test. Deze verbinding activeert Autophagy. Fase 1/2.

In vitro

FK866 (Daporinad) induceert bij lage concentraties variërend van 0,09-27 nM dosisafhankelijke cytotoxiciteit in 41 hematologische kwaadaardige cellen, waaronder acute myeloïde leukemie [AML], acute lymfoblastische leukemie [ALL], mantelcellymfoom [MCL], chronische lymfatische leukemie [CLL] en T-cellymfoom. Bij concentraties variërend van 0-10 nM induceert deze verbinding celdood, een effect dat onafhankelijk is van caspase-activering maar geassocieerd is met depolarisatie van het mitochondriale membraan. Het induceert ook dosisafhankelijk uitputting van intracellulaire NAD- en ATP-gehalten en celdood in verschillende hematologische kankercellen bij concentraties variërend van 0-10 nM. Bij een concentratie van 10 nM remt het de PBEF-geïnduceerde secretie van MMP-3, CCL2 en CXCL8 in HFFF2-cellen.

In vivo

FK866 (Daporinad), intraperitoneaal toegediend in een dosis van 20 mg/kg tweemaal daags gedurende 4 dagen, wekelijks herhaald gedurende 3 weken, voorkomt en annuleert tumorgroei in C.B.-17 SCID muizen xenograftmodellen van humane AML, lymfoblastisch lymfoom en leukemie. Bij een dosis van 0,12 mg/kg/uur voorkomt deze verbinding gewrichtsdestructie en leukocyteninfiltratie door remming van PBEF bij muizen met CIA. }

FK866 (Daporinad) is added in 50 μL of culture medium, with culture medium alone serving as control. After 72 or 96 hours of incubation, 15 μL of dye solution is added to each well and cells are incubated for an additional 4 hours. Stop solution (100 μL/well) is added for 1 hour and the absorbance is read at 570 nm on a spectrophotometer. For trypan blue dye exclusion staining, 0.5 × 10⁵ cells/well is grown in 6-well plates with 1 mL media in the absence or presence of this compound for 96 hours. Cells from each sample are incubated with 10 μL trypan blue solution (at a 1:1 ratio [vol/vol] for 1 minute). Cell survival is determined by calculating proportion of live (unstained) cells.
