Beschrijving FB23-2 is een potente en selectieve FTO-remmer die direct aan FTO bindt en selectief de N6-methyladenosine (m6A) demethylase-activiteit van deze verbinding remt met een IC50 van 2,6 μM.

In vitro

FB23-2 bindt direct aan FTO en remt selectief de m6A-demethylase-activiteit. Door FTO-depletie na te bootsen, onderdrukt deze verbinding dramatisch de proliferatie en bevordert de differentiatie/apoptose van cellen van menselijke acute myeloïde leukemie (AML) cellijnen en primaire blast-AML-cellen in vitro.
In vivo

FB23-2 remt significant de progressie van menselijke AML-cellijnen en primaire cellen in xenotransplanteerde muizen.

    NB4 and MONOMAC6 cells are treated with DMSO or FB23-2 at varying concentrations for 72 h, while MA9 and FLT3/NPM1 primary cells isolated from AML mice, five human AML cell (MA9.3ITD, MA9.3RAS, U937, ML2, and MV4-11), and human primary AML cells are treated with DMSO or 5 μM of this compound for 72 h for dot blot assay.
