Beschrijving CY-09 is een specifieke NLRP3 inflammasoom-remmer die NLRP3 zelf direct target. Het is getest tegen de vijf belangrijkste cytochroom P450-enzymen 1A2, 2C9, 2C19, 2D6 en 3A4 met half maximale remmende concentratie (IC50) waarden van respectievelijk 18,9, 8,18, >50, >50 en 26,0 µM.

CY-09 blokkeert specifiek de NLRP3-activering in macrofagen. Deze verbinding remt de NLRP3-oligomerisatie en de inflammasoomassemblage. Het bindt direct aan NLRP3 en remt de ATPase-activiteit. De metabolische stabiliteit van deze chemische stof werd eerst geëvalueerd met behulp van microsomen van menselijke en muizenlever, waarbij een gunstige stabiliteit werd vertoond met een halfwaardetijd van >145 min voor zowel menselijke als muizenmicrosomen. In vivo

CY-09 remt de NLRP3-activering in vivo en voorkomt neonatale lethaliteit in een muismodel van CAPS. In farmacokinetische studies die werden geëvalueerd bij C57BL/6J-muizen waaraan een enkele i.v. of orale dosis werd toegediend, vertoont dit middel gunstige farmacokinetische eigenschappen, met een halfwaardetijd van 2,4 uur, een oppervlakte onder de curve van 8.232 (uur·ng)/ml en een biologische beschikbaarheid van 72%. Het keert metabole stoornissen bij diabetische muizen om door remming van NLRP3-afhankelijke ontsteking. Deze chemische behandeling heeft opmerkelijke gunstige effecten op meta-inflammatie, hyperglycemie en insulineresistentie bij diabetische muizen.

    5 × 105/ml BMDMs and 6 × 106/ml PBMCs were plated in 12-well plates. The following morning, the medium was replaced, and cells were stimulated with 50 ng/ml LPS or 400 ng/ml Pam3CSK4 (for noncanonical inflammasome activation) for 3 h. After that, CY-09 or other inhibitors were added into the culture for another 30 min, and then the cells were stimulated for 4 h with MSU (150 µg/ml), Salmonella typhimurium (multiplicity of infection) or for 30 min with ATP (2.5 mM) or nigericin (10 µM). Cells were transfected with poly(dA:dT) (0.5 µg/ml) for 4 h or LPS (500 ng/ml) overnight. Cell extracts and precipitated supernatants were analyzed by immunoblot.
