Beschrijving CPI-203 is een potente BET bromodomain remmer met een IC50 van 37 nM voor BRD4.

In vitro

CPI203 remt BRD4 in vitro en in cellen, terwijl het de BRD4 kinaseactiviteit in vitro niet beïnvloedt. CPI203 oefent een cytostatisch effect uit in alle 9 geanalyseerde MCL-cellijnen met een GI50 variërend van 0,06 tot 0,71 μM, met lage cytotoxiciteit in normale PBMCs van gezonde donoren. Bovendien activeert CPI203, door IRF4 en MYC te targeten, efficiënt het celdoodprogramma in MCL-cellen.

In vivo

BRD4 medieert CTD Ser2-fosforylering in vivo. Bij REC-1 tumor-dragende muizen versterkt de combinatie van CPI203 (2,5 mg/kg i.p.) synergetisch de antitumorale eigenschappen van elk enkel middel via de opheffing van MYC- en IRF4-expressie en de inductie van apoptose.

  • Methode

    MCL primary cells and cell lines are incubated as indicated with CPI203. MTT is added for 2-6 additional hours before spectrophotometric measurement. Each measurement is made in triplicate. Values are represented using untreated control cells. The GI50 is calculated as the concentration that produced 50 % growth inhibition. Combination indexes (CIs) are calculated by using the Calcusyn software version 2.0. The interaction between two drugs is considered synergistic when CI <1.
