Beschrijving Colivelin (CLN) is een hersen-penetrerend neuroprotectief peptide met krachtige langetermijncapaciteit tegen Aβ-depositie, neuronale apoptose en synaptische plasticiteitsdefecten bij neurodegeneratieve ziekten. Colivelin is een activator van STAT3.

Colivelin, een hersenpenetrerend neuroprotectief peptide, verhoogt significant de p-STAT3 eiwitniveaus in BV-2 cellen.
In vivo

Deze verbinding beschermt tegen ischemisch hersenletsel en verbetert neurologische uitkomsten, door de opregulatie van anti-apoptotische genen te verhogen en de JAK/STAT3 signalering te activeren na een ischemische beroerte.

  • Methode

    BV-2 cells are maintained at 37℃ with 5% CO2. To build OGD models, BV-2 cells are washed with PBS for 3 times, and cultured in glucose-free DMEN and placed in a hypoxia incubation chamber filled with the gas mixture for 4 h at 37℃. The culture medium is then replaced by the normal medium containing 4.5 g/L glucose and cells are cultured under normoxic conditions for reoxygenation for 24 h. BV-2 cells are treated with 50 µg/ml rmMFG-E8 or/and this compound, respectively. The expression changes of proteins (p-STAT3 and SOCS3) are analyzed by Western blot assay.
