Biologische activiteit

Beschrijving CGP 57380 is een potente MNK1-remmer met een IC50 van 2,2 μM, die geen remmende activiteit vertoont op p38, JNK1, ERK1 en -2, PKC, of c-Src-achtige kinasen. Deze verbinding reguleert β-catenine op en potentieert stralingsgeïnduceerde apoptose.

In vitro CGP57380 remt de fosforylering van eIF4E in cellulaire assays met een IC50 van ongeveer 3 μM. Deze verbinding veroorzaakt defosforylering van eIF4E en induceert een verdere toename van de cap-afhankelijke reporter in 293-cellen. Dit resulteert in dosisafhankelijke afnames in Ang II-gestimuleerde fosforylering van eIF4E, eiwitsynthese en VSMC-hypertrofie. Deze chemische stof sensibiliseert wildtypecellen voor serumonttrekkingsgeïnduceerde apoptose in muizenembryofibroblasten (MEF's). Het voorkomt de seriële herplatingsfunctie van BC-voorlopercellen.
In vivo CGP57380 (40 mg/kg/dag i.p.) dooft krachtig het vermogen van BC CML-cellen uit om immuundeficiënte muizen serieel te transplanteren en te functioneren als LSC's.

Klantproductvalidatie

PC-3 cells were seeded in 6-well plates. On the next day, cells were treated with the indicated concentrations of 3-AWA, LY294002, CGP57380 or Rapamycin alone and or combination for 12 h. The cells were then subjected to preparation of whole-cell lysates for detection of the indicated proteins using western blotting. The western blots are the representative of three similar experiments.
