Biologische activiteit

Beschrijving Carboplatin, een derivaat van CDDP, is een DNA synthesis inhibitor die bindt aan DNA, replicatie en transcriptie remt en celdood induceert en interfereert met het celreparatiemechanisme in A2780, SKOV-3, IGROV-1 en HX62 cellen. Het induceert de vorming van DNA interstrand crosslinks en DNA-eiwit crosslinks. Oplossingen zijn onstabiel en moeten vers worden bereid. DMSO wordt niet aanbevolen om op platina gebaseerde geneesmiddelen op te lossen, wat gemakkelijk kan leiden tot inactivering van het geneesmiddel.

In vitro

Carboplatin vertoont een remmend effect op celproliferatie in een panel van menselijke eierstokkankercellijnen, waaronder A2780, SKOV3 en IGROV-1 cellen met respectievelijk IC50's van 6,1 μM, 12,4 μM en 2,2 μM. Deze verbinding vertoont ook antiproliferatieve activiteiten in longcarcinoïde cellijnen, zoals UMC-11, H727 en H835 cellen met respectievelijk IC50's van 36,4 μM, 3,4 μM en 35,8 μM.

In vivo

In A2780 tumor xenografts toont Carboplatin (60 mg/kg via i.p.), toegediend als enkelvoudige middelen, een bescheiden antitumor effect met relatieve tumorvolumes op dag 6 van 8,4 vergeleken met de controle van 11,9, en de tumorvolumes op dag 6 relatief ten opzichte van controle (T/C) van 67%. Voor de VC8 (Brca2-deficiënte) xenografts vertraagt deze verbinding de tumorgroei en vermindert de tumormassa met 42% vergeleken met de vehikelgroep.

Kenmerken Dit product wordt niet aanbevolen om op te lossen in dimethylsulfoxide (DMSO).

    3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) assays: Exponentially growing A2780, SKOV3, IGROV-1 and HX62 ovarian cancer cells are plated in 96 well plates. A range of drug concentrations are added and the plates are incubated for 72 hours to allow for 3–4 doubling times. Each experiment is carried out in triplicate. Sulforhodamine B (SRB) assays: Exponentially growing A2780 cells are plated in 96 well microtitre plates. For experiments studying concomitant exposure, cells are exposed to increasing concentrations of both drugs for 96 hours. For experiments studying the effect of sequence of exposure to 17-AAG or this compound cells are exposed to increasing concentrations of 17-AAG or this chemical for 24 hours. A period of 24-hour exposure to the first agent is chosen so that the A2780 cells would be exposed to the first drug for at least one doubling time (18-24 hours). The cells are then washed with sterile phosphate buffered saline and the medium is replenished. Following this, the second drug (to which the cells are not exposed to in the first 24 hours) or medium is added for 96 hours. All experiments are carried out in triplicate. The results of combination studies are analyzed using the well-established principles of median effect analysis method. The effects of the combination are calculated using an in-house spreadsheet.

<p>Detection of apoptotic responses was found to increase in epimorphin-treated  A1847 with increasing concentrations of carboplatin compared to those of the untreated controls. Data were normalized to the controls and are represented as means ± S.D. [*p<0.05 compared with control]. Images of apoptotic cells were captured at 10X magnification, with at least 3 images per well, using an upright phase-contrast microscope (T3.15A; Fisher Scientific).</p>
