 3.600mg/ml (9.93mM) Taking the 1 mL working solution as an example, add 50 μL of 72 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Biologische activiteit

Beschrijving Calpeptin is een krachtige, celpermeabele calpain-remmer met een ID50 van respectievelijk 52 nM, 34 nM, 138 nM en 40 nM voor Calpain I (varkenserythrocyten), Calpain II (varkensnier), Papainb en Calpain I (menselijke bloedplaatjes). Deze verbinding vermindert apoptosis en intracellulaire ontstekingsveranderingen in spiercellen.

In vitro Calpeptin remt 20K-fosforylering op een dosisgerelateerde manier in bloedplaatjes gestimuleerd door trombine, ionomycine of collageen. In differentiërende PC12-cellen bevordert deze verbinding de neurietverlenging via remming van Calpain-activiteit. In retinale ganglioncellen van ratten vermindert het apoptosis, handhaaft het een normaal celmembraanpotentiaal en biedt het zo functionele neuroprotectie.
In vivo In een feline rechterventrikel (RV) PO (RVPO) model blokkeert calpeptin (0,6 mg/kg, i.v.) de activering van calpaïne en caspase-3, de splitsing van hun substraten en geprogrammeerde celdood van cardiomyocyten. In een rattenmodel voor focale cerebrale ischemie-reperfusieschade vermindert deze verbinding de neuronale apoptosis in de hippocampale CA1-sector via remming van de expressie van Caspase-3.

Klantproductvalidatie

Western blotting detection of focal adhesion kinase (FAK) expression and its cleaved fragments with different treatments. DTX represents docetaxel, CPP represents calpeptin (a calpain inhibitor). The experiments were carried out 3 times, obtaining essentially similar results.
