Biologische activiteit

Beschrijving	Calcipotriene (MC903) is een synthetisch derivaat van calcitriol, een vorm van Vitamin D. Het kan differentiatie induceren en de proliferatie van keratinocyten onderdrukken, waardoor abnormale keratinocytveranderingen bij psoriasis worden omgekeerd en normalisatie van de epidermale groei wordt bewerkstelligd. Calcipotriol is een synthetisch VitD3-analoog met een hoge affiniteit voor de Vitamin D-receptor.Oplossingen zijn onstabiel en dienen vers te worden bereid.
In vitro	Calcipotriene verhoogt de IL-10-secretie in psoriatische huid en in humane T-cellen, induceert de secretie van type 2-cytokines door T-cellen en moduleert significant de differentiatie van DC's en T-cellen.
In vivo	Calcipotriol onderdrukt de ontwikkeling van huidkanker bij muizen op een TSLP-afhankelijke manier. Calcipotriol is een goed verdragen topische medicatie voor psoriasis. De lage concentratie calcipotriol in de klinisch beschikbare formulering (0,005% zalf) vermindert het effect tegen de ontwikkeling van huidkanker bij muizen.
