 0.500mg/ml (1.02mM) Taking the 1 mL working solution as an example, add 50 μL of 10 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify it; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Biologische activiteit

Beschrijving CA3 (CIL56) heeft krachtige remmende effecten op de YAP1/Tead transcriptionele activiteit en richt zich primair op YAP1-hoge en therapieresistente slokdarmadenocarcinoomcellen met CSC-eigenschappen. CA3(CIL56) induceert Ferroptosis en ijzerafhankelijke reactieve zuurstofspecies (ROS).

In vitro

CA3 (CIL56) remt de groei van slokdarmadenocarcinoomcellen in vitro sterk. Het kan de proliferatie van tumorcellen effectief onderdrukken, apoptose induceren, de vorming van tumorsferen en de populatie van ALDH1+-cellen verminderen. Deze verbinding remt specifiek de Tead/YAP1 transcriptionele activiteit, maar vertoont geen remmende activiteit op andere transcriptionele factoren - Super-TOP/Wnt, CBF1/Notch en AP-1 na cotransfectie van hun respectieve individuele promoter luciferases in 293T-cellen. CA3 remt bij voorkeur CSC-eigenschappen die zijn verrijkt in stralingsresistente slokdarmadenocarcinoomcellen.
In vivo

CA3 (CIL56) oefent sterke antitumoractiviteit uit in een xenograftmodel zonder duidelijke toxiciteit.

  • Methode

    SKGT-4 and JHESO cells are seeded onto 6-well plates (1 × 105/well) in DMEM and cultured for 24 hours to allow for cell attachment. The cells are then treated with 0.1% DMSO (control) or CA3 (CIL56) at different doses as indicated for 48 hours. Next, the cells are harvested, fixed with methanol, washed, treated with RNase A, and stained for DNA with propidium iodide, and their DNA histograms and cell-cycle phase distributions are analyzed using flow cytometry.
