 0.500mg/ml (1.33mM) Taking the 1 mL working solution as an example, add 50 μL of 10 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify it; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Beschrijving Brequinar is een remmer van dihydroorotaat dehydrogenase (DHODH) met een IC50 van ~20 nM in vitro.

Brequinar natrium (BQR) vermindert de PyN-niveaus in Con A-gestimuleerde T-cellen en remt de celproliferatie bij lage concentraties, wat kan worden omgekeerd door uridine. Uridine is echter niet in staat om de effecten van hoge concentraties BQR om te keren. BQR bezit het vermogen om tyrosinefosforylering te remmen.
In vivo

Brequinar natrium (BQR) induceert anemie door remming van de DHO-DHase activiteit en de novo pyrimidinesynthese in vivo. Gelijktijdige toediening van BQR met uridine voorkomt anemie.

    Lymphocytes are isolated from BALB/c spleen by centrifugation through a Ficol gradient and are depleted of macrophages and B cells by adherence to goat anti-mouse IgG-coated plates. The nonadherent cells are harvested and cultured at 4 to 5× 105/ml in RPMI 1640 supplemented with 10% FBS. Stimulation is performed with 5 μg/ml Con A in the presence of indicated concentrations of Brequinar or uridine. After 40 to 66 h, the cells are harvested and the levels of cell proliferation or purine NTP and PyNTP are quantified.
