Beschrijving

Bobcat339 is een selectieve cytosine-gebaseerde TET enzyme remmer met IC50 van respectievelijk 33 μM en 73 μM voor TET1 en TET2. Bobcat339 kan de DNA 5-hydroxymethylcytosine abundantie verminderen door de TET-enzymfunctie in levende cellen te remmen en ondersteuning bieden voor het nut ervan als een levensvatbare farmacologische probe.

Bobcat339, de TET2 enzymactiviteitsremmer, verlaagt de TET2 enzymactiviteit en beschermt cellen tegen neurotoxine schade in SH-SY5Y cellen.

SH-SY5Y cells are seeded into 96-well plates at a density of 5,000 cells/well with 100 μl of cell medium in each well. Bobcat339 (3 μM) is preincubated with cells for 12 h before MPP+ (2.5 mM) treatment. Then, 10 μl of CCK-8 working solution is added to each well to incubate for about 1 h at 37 C. The wells containing only the culture medium served as blanks. The absorbance value at 450 nm is measured.
