BMS-582949 (PS540446) is een potente en selectieve p38 mitogen-activated protein kinase (p38 MAPK)-remmer met een IC50 van 13nM, die zowel de p38 kinase-activiteit als de activering van p38 remt.

In vitro

BMS-582949 blijkt de p38-activering in cellen te remmen, gemeten aan de hand van de fosforylering van p38. Behandeling van cellen waarin p38 door LPS was geactiveerd met deze verbinding keerde de p38-activering snel om, zoals aangetoond door het verlies van fosforylering van p38. Het is daarom een dubbelwerkende p38 kinase-remmer, die zowel de p38 kinase-activiteit als de p38-activering in cellen remt. De binding aan p38a resulteert in een conformationele verandering van de activeringslus die wordt gefosforyleerd door upstream kinasen, daarom remt het de fosforylering van p38 door upstream MKK door een minder toegankelijke conformatie van de activeringslus te induceren.

In vivo

De klaring van BMS-582949 bij muizen is 4,4 mL/min/kg. En bij een orale dosis van 10 mg/kg is de AUC0-8 h bij muizen voor deze verbinding 75,5 μM•h. Deze chemische stof vertoonde orale biologische beschikbaarheidswaarden van respectievelijk 90% en 60% bij muizen en ratten.
