Beschrijving BLU9931 is een potente, selectieve en irreversibele FGFR4-remmer met een IC50 van 3 nM, ongeveer 297-, 184- en 50-voudige selectiviteit ten opzichte van FGFR1/2/3, respectievelijk.

In vitro In MDA-MB-453-cellen remt BLU9931 potent de fosforylering van de FGFR4-signaalroute. Deze verbinding remt de proliferatie van HCC-cellijnen die een intact FGFR4-signaalcomplex tot expressie brengen, zoals Hep 3B-, HUH-7- en JHH-7-cellijnen, met een EC50 van <1 μM. Het remt ook de proliferatie in PDX-afgeleide cellijnen met een intacte FGFR4-signaalroute.
In vivo Bij muizen met FGF19-geamplificeerde Hep 3B-levertumoren leidt BLU9931 (300 mg/kg, p.o.) tot tumorregressie en voorkomt dit gewichtsverlies geïnduceerd door tumoren. Bij muizen met FGF19-overexpressieve PDX-afgeleide LIXC012-xenografts leidt behandeling met deze verbinding ook tot tumorregressie.

  • FGFR1-4 Biochemische assays

    FGFR-kinase-inhibitie-assays worden uitgevoerd bij KM voor ATP. Picomolaire tot lage nanomolaire concentraties van FGFR-eiwitten worden geïncubeerd in 1× Kinase Reactie Buffer (KRB) met 1 μM CSKtide en 50 tot 250 μM ATP bij 25°C gedurende 90 minuten in aanwezigheid of afwezigheid van een gedoseerde concentratieserie van deze verbinding. Alle reacties worden beëindigd door toevoeging van Stopbuffer, en platen worden afgelezen op een Caliper EZReader2. IC50-waarden worden gefit met een vierparameter log[Inhibitor] versus responsmodel met zwevende Hill Slope.

    Established and PDX-derived HCC cell lines are seeded in 96-well plates in respective growth media, allowed to attach overnight, and treated with a dilution series of test compounds for two cell-doubling times. Cell viability is determined by CellTiter-Glo, and results represented as background-subtracted relative light units normalized to a DMSO-treated control. Relative EC50 values are determined at 50% inhibition between the top and bottom plateau of the dose–response curve.
