Beschrijving

Apcin (APC-remmer) is een remmer van de E3 Ligase-activiteit van het mitotische anaphase-promoting complex/cyclosome (APC/C) dat bindt aan Cdc20 en de ubiquitilatie van D-box-bevattende substraten competitief remt.

Apcin is een nieuw celdoorlaatbaar molecuul dat de interactie tussen APC/C en Cdc20 blokkeert. Dit middel remt de groei van OS-cellen en induceert significante apoptose. De invasie- en mobiele vermogens van OS-cellen worden ook aanzienlijk onderdrukt door deze chemische behandeling. Bovendien worden Bim en p21 opgereguleerd in OS-cellen na deze behandeling. De remming van CDC20 met zijn remmer Apcin vermindert de adhesiesnelheid en proliferatiecapaciteit van respectievelijk RL95-2 en HEC-1A cellen.

CDC20-remmer Apcin remt embryo-implantatie in vivo.

Protocol (uit referentie)

Celassay:^{^[2]}	Cellijnen Human osteosarcoma cell lines MG63 and U2OS Concentraties 25 μM, 50 μM, 75 μM Incubatietijd 24 h, 48 h, 72 h Methode MG63 and U2OS (2.5×10³ cells/well) are seeded in 96‑well plates and cultured overnight. Different concentrations of apcin are added to the medium and cultured for 24, 48 and 72 h, respectively. An MTT assay is then carried out to assess the cell viability according to the manufacturer's protocols. Then, 10 μl of MTT solution (0.5 mg/ml) is added to each well followed by incubation for 4 h. The liquid supernatant is then drained off and 100 μl of DMSO is added to each well to dissolve the crystals. The absorbance of each well at 490 nm is determined using a Multimode Reader of SpectraMax M5.
