Beschrijving AGI-6780 is een krachtige en selectieve remmer van de IDH2 R140Q-mutant met een IC50 van 23 nM.

In vitro AGI-6780 vermindert potent de (R)-2-hydroxyglutarate (2HG)-niveaus in cellijnen die ectopisch IDH2/R140Q overexpressen met een EC50 van 20 nM, met uitstekende selectiviteit tegenover andere dehydrogenases. Deze verbinding keert het IDH2/R140Q-geïnduceerde differentiatieblok in TF-1-cellen om, en induceert blastdifferentiatie in primaire menselijke IDH2/R140Q AML-patiëntmonsters.

Western blot of KMM-1 and NCI-H929 cells, UT, treated with DMSO, AGI-6780 (KMM-1: 5 μM; NCI-H929: 10 μM), CFZ (KMM-1: 5 nM; NCI-H929: 2.5 nM), or a combination of the 2 drugs. Cell lysates were immunoblotted using the indicated antibodies 24 hours posttreatment. Vinculin protein expression was included for protein loading normalization.
