Beschrijving

Adagrasib (MRTX849) is een potente, selectieve en covalente KRASG12C-remmer die gunstige, geneesmiddelachtige eigenschappen vertoont. Het modificeert selectief het gemuteerde cysteïne 12 in GDP-gebonden KRASG12C en remt KRAS-afhankelijke signalering.

In vitro

Om de breedte van de Adagrasib (MRTX849)-activiteit te evalueren, wordt het effect ervan op de cellevensvatbaarheid bepaald over een panel van 17 KRAS^G12C-mutante en drie niet-KRAS^G12C-mutante kankercellijnen met behulp van 2D (3-daagse, adherente cellen) en 3D (12-daagse, sferoïden) celgroeicondities.
