Biologische activiteit

Beschrijving Actinomycin D (Dactinomycin, Act D, RASP-101) is een belangrijk polypeptide-antibioticum geïsoleerd uit bodembacteriën van het geslacht Streptomyces. Actinomycin D (Dactinomycin) remt DNA repair en stopt de cell cycle in de G1-fase met een IC50 van respectievelijk 0,42 μM en 0,4 nM. Actinomycin D is een RNA- en eiwitsyntheseremmer die bacteriële eiwitsynthese kan onderdrukken. Actinomycin D is een autophagy-activator, induceert p53-onafhankelijke celdood en verlengt de overleving bij hoogrisico chronische lymfatische leukemie.Dit product is een gevaarlijke chemische stof (acute toxiciteit/ontvlambaar/huidcorrosief). Gebruik het met een beschermend gezichtsmasker, handschoenen en kleding.

In vitro

Actinomycin D (Dactinomycin) vermindert de SMC-proliferatie aanzienlijk via de remming van BrdU-incorporatie bij 80 nM. Dit wordt verder ondersteund door de G1-fase-arrestatie met behulp van een flowcytometrische analyse. De eiwitexpressieniveaus van prolifereerd cell nuclear antigen (PCNA), focal adhesion kinase (FAK) en Raf worden allemaal onderdrukt door deze verbinding. Extracellulaire signaal-gereguleerde kinasen (Erk) die betrokken zijn bij de celcyclus-arrestatie blijken erdoor te worden verhoogd.
In vivo

Actinomycin D (Dactinomycin) richt zich op overlevingseiwitten TOSO, BCL2 en MCL1 en is actief in twee verschillende muismodellen die gekenmerkt worden door een ongemuteerde B-celreceptor of inactieve p53-functie, beide bekende negatieve prognostische factoren bij CLL.

    Cultured SMC are starved for 24 h followed by incubation with various doses of actinomycin D (dactinomycin) at 37℃. Drug treatment is carried out for 18-24 h. Since it is dissolved in 0.1% DMSO, a vehicle control containing DMSO is also included.
